MAP / IMPACT
IMPACT PROFILES - AN ALTERNATIVE FOR MAP TESTING Why test?: Contamination of biological specimens, such as cell lines, hybridomas and tumor cells, with rodent pathogens can result in devastating outbreaks of disease in laboratory animals implanted with these materials and confounding and deleterious effects on tissue culture-based experiments. PCR-based alternative to MAP testing. Infectious Microbe PCR Amplification Test or IMPACT was developed and is performed at RADIL University of Missouri. All PCR assays have been extensively validated. Comparison of IMPACT results with MAP testing results for representative DNA and RNA viruses indicated that the sensitivity of the IMPACT was equal or greater than that of MAP testing. Turn around time for IMPACT is usually 7-10 business days. Compared to 6-8 weeks for MAP testing. This type of testing needs to be arranged for in advance with ACP Veterinary Diagnostic Lab. A minimum of 72 hours notice is required to accommodate the need for Dry ice shipment of the samples to the reference. There is a $30.00 charge for dry ice shipping. (Shipping charge can be avoided by timing your shipment to go out with our regular shipment however, samples must be in our lab no later than 4:00PM on Wednesday). Samples are shipped on dry ice with an overnight courier. An ACP Diagnostic Lab Request form should be completed to increase efficiency of shipping, result distribution and billing. Information concerning the origin of the cells contained in the sample is also required and should be included on the form in the history section. Contact us at 534-4264 or 534-7173 Sample requirements for IMPACT testing: 2 vials of each sample with a minimum of 1x10 to the 7th power cells / vial frozen (in about 2mls of culture media) and transported on dry ice to the ACP Veterinary Diagnostic Lab located in BSB room B200. Pooling of cell lines is permitted (3 lines maximum). Down sides of pooling: the tests are not validated for pooled samples though it is not uncommon; positive results on pooled samples would require re-testing of lines pooled separately to determine which line or lines are contaminated. There are four different profiles available but IMPACT PROFILE II is the Profile ACP Veterinary Staff recommends. **Profile IV must be cleared by Dr. Jenne
